E’ noto che nei vertebrati omeotermi gli anestetici e la chirurgia alterano la termoregolazione, inducendo ipotermia. Questo studio mira a valutare l'effetto della anestesia generale e dell'intervento chirurgico sulla temperatura corporea in cani sottoposti a orchiectomia, valutando l'interazione tra TSH, ormoni tiroidei e cortisolo. Sette cani, di età compresa tra i 3-5 anni e con peso corporeo medio 15-20 kg , in buone condizioni di salute, sono stati premedicati con atropina (0,04 mg / kg / IM) e xilazina (0,55 mg / kg / IM) e 10 minuti dopo indotti all’anestesia generale con ketamina (0,02 mg / kg / iv). L'anestesia generale è stata mantenuta con isoflurano al 2-3 % in circuito chiuso e con un flusso di ossigeno di 10 ml / kg. L’orchiectomia è stata effettuata su tutti i cani dallo stesso staff chirurgico, nella stessa mattinata, tra le 7:30 e le 13:30, così come il prelievo di sangue (dalla vena cefalica) e il rilievo( per via rettale) della temperatura corporea. Il campionamento di sangue e il rilievo della T° sono stati effettuati: poco prima della premedicazione (T0), 1 ora dopo (che corrisponde alla fase di risveglio, T1) e dopo 2 ore T0 (T2). La T° è diminuita mediamente da 38,7 ± 0,1 (a T0); a 37,7 ± 0,1 (a T1), per poi risalire di nuovo a 38,3 ± 0,1 (a T2). TSH, FreeT3 e FreeT4 sono stati analizzati mediante ELISA Canine kit (Endocrinetech, Newark, Canada); il cortisolo è stato analizzato mediante un kit umano (Intermedical S.r.l. Italia). Il livello ormonali nei tre tempi considerati sono stati: TSH (pg / ml) 1,04 ± 0,91; 0,86 ± 0,61; 0,81 ± 0,66; fT3 (pg / ml) 1,28 ± 0,77; 1,24 ± 0,94; 1,18 ± 1,04; fT4 (pg / ml) 1,21 ± 0,51; 1,09 ± 0,49; 1,27 ± 0,46; il cortisolo (ng / ml) 11,9 ± 4,52; 19,3 ± 11,1; 36,7 ± 27,9 ;. A T2, in relazione alla diminuzione della T ° C, si verifica una diminuzione dei livelli di TSH, fT3 (P <0.001) e fT4. Tra TSH e fT3 (R = 0,952; p <0.001) a T2 si è osservata una correlazione positiva (test di Pearson). Il cortisolo è aumentato significativamente (P <0.05) già a T0 (11,9 ± 4,52) a T1 (19,3 ± 11,1) e T2 (36,7 ± 27,9). Gli anestetici utilizzati e l’intervento di orchiectomia influenzano la termoregolazione nei cani;la condizione di stress (che si traduce in alti livelli di cortisolo) è già presente prima della premedicazione.

It’s known that in homoeothermic vertebrates the anesthetics and the surgery alter thermoregulation, inducing hypothermia. This study aims at assessing the effect of the general anesthesia and of the surgical operation on body temperature in dogs undergoing orchiectomy, evaluating the interaction between TSH, thyroidal hormones and cortisol. Seven dogs, 3-5 yo and 10-25 kg/bw, in good health condition, were premedicated with Atropine (0.04 mg/kg/im) and Xilazine (0.55 mg/kg/im) and injected, 10 min later, with Ketamine (0.02 mg/kg/iv), to induce anesthesia. The general anesthesia was maintained with 2-3% Isoflurane in 10 ml/kg Oxygen flow in closed circuit system. Orchiectomy was performed in all dogs from the same staff in the same day.The rectal temperature and blood samples (from cephalic vein) were collected: just before the premedication (T0), 1 hour after recovery from anesthesia (T1) and after 2 hours from the premedication (T2).The body temperature decreased from 38.7±0.1 (at T0) to 37.7±0.1 (at T1) than raised again to 38.3±0.1 (at T2). TSH, FreeT3 and FreeT4 were analysed by ELISA Canine kits(Endocrinetech, Newark, Canada); Cortisol was analysed by an human kit (InterMedical S.r.l. Italia). The level of hormones in the 3 considered times were: TSH (pg/ml) 1.04±0,91;0.86±0,61; 0.81±0,66; fT3 (pg/ml) 1.28±0,77; 1.24±0,94; 1.18±1,04; fT4 (pg/ml) 1.21±0,51;1.09±0,49;1.27±0,46; Cortisol (ng/ml) 11.9±4,52;19.3±11,1; 36.7±27,9;. At T2, in relation to the decrease of the T°C, a lowering of the levels of TSH, fT3 (P<0.001) and fT4, was observed. Between TSH and fT3 (R=0.952; P<0.001) at T2 a positive correlation (Pearson’s Test), was found. The cortisol increased significantly (P<0.05) from T0 (11,9 ± 4,52) to T1 (19,3 ± 11,1) and T2 (36,7 ± 27,9). Anesthetics and orchiectomy affect thermoregulation in dogs ; the condition of stress (high cortisol levels) is already present before the premedication.
